A thin section of wood cells was used for this investigation. The spectra were recorded with a 100X water immersion objective (NA = 1) with a small confocal aperture enabling a spatial resolution at the diffraction limit of incident light. For the map 200 x 200 points with 0.5 µm spacing were recorded. The acquisition time for single spectra was 500 ms each. Raman image was created by using integrals of three characteristic bands representing the different cell components.
