3D Confocal Raman Microscopy for Detection of Minor Crystalline API Form in a Pharmaceutical Extrudate

This application note demonstrates the use of 3D confocal Raman microscopy for the characterization of pharmaceutical extrudates, with a focus on detecting and distinguishing crystalline and amorphous forms of an active pharmaceutical ingredient (API) within a complex matrix. Using the LabRAM Soleil™ Raman microscope, high spatial and spectral resolution Raman measurements were performed to map the API distribution in three dimensions. The confocal approach allows precise localization of minor crystalline phases embedded in the extrudate, providing critical insight into solid-state heterogeneity. This non-destructive methodology supports efficient quality control and formulation development, helping to ensure product performance and regulatory compliance in pharmaceutical manufacturing.
