Environmental pollutants such as ozone generate oxidative stress that alters the molecular composition of the skin. This study demonstrates a multimodal in vivo spectroscopic workflow combining FT-IR and Confocal Raman spectroscopy (RZ660) for the non-invasive characterization of ozone-induced skin damage. Controlled ozone exposure produced measurable oxidative modifications within 15 minutes, detected at the skin surface by FT-IR and up to 10 µm depth by Raman spectroscopy. Time-resolved measurements revealed the evolution and recovery of these molecular alterations over 24 hours. The complementary information provided by both techniques highlights their capability to deliver quantitative, depth-resolved biomarkers of oxidative stress. This approach establishes vibrational spectroscopy as a powerful platform for studying environmental impacts on biological tissues and monitoring dynamic molecular changes in vivo.
