Nanoparticle Tracking Analysis (NTA) has been extensively used for extracellular vesicle (EV) size and quantification. Nonetheless, there exists a consensus within the EV community that NTA is merely a “quick-and-dirty” screening tool, characterized by a “reproducibility crisis” (as described at the ISEVxTECH meeting in 2022). The heterogeneity of EV's as well as the size of large-EVs present a significant challenge to measurement accuracy. This is also true for exosome, regardless of its biogenesis.
We challenge this thinking and invite you to leverage HORIBA’s ViewSizer® 3000 simultaneous Multi-Laser NTA that was first developed and patented by the University of California, San Diego for oceanography applications (who also asked the same questions above), in your EV characterizations.

In this talk, we discuss the topic of EV isolation with a focus on the most popular method of size exclusion chromatography and the challenges in characterizing their size distribution. Additionally, Ya-Hong Xie from UCLA will share his perspective of how this line of research could lead to deepening our understanding of exosomal biology pertaining to their diagnostic and therapeutic promises.
