Fluorescence Spectroscopy

HORIBA’s Fluorescence division incorporates technology from SPEX, IBH, Jobin-Yvon, PTI and SLM - the dominant names in Fluorescence Spectroscopy.
What can fluorescence lifetime reveal that fluorescence intensity alone cannot?
In this educational webinar, discover the fundamentals of fluorescence lifetime and time-resolved fluorescence spectroscopy, and learn how these techniques provide information about excited-state dynamics, molecular environments and photophysical processes.
The webinar introduces the principles of fluorescence lifetime and explains how Time-Correlated Single Photon Counting (TCSPC) is used to measure fluorescence decays from picoseconds to nanoseconds. Explore the key components of a lifetime measurement system, including pulsed excitation sources, timing electronics and single-photon detectors, as well as the principles of decay fitting and data analysis.
The session also introduces Time-Resolved Emission Spectroscopy (TRES) and shows how spectral and temporal information can be combined to distinguish emitting species with overlapping spectra.
Finally, discover techniques for measuring longer-lived phosphorescence, including Multi-Channel Scaling (MCS), Boxcar and Single-Shot Transient Digitizer (SSTD) methods.
Application examples from life sciences and materials science demonstrate how fluorescence lifetime analysis can investigate proteins, molecular interactions, quantum dots, perovskites, lanthanides and other luminescent materials.

Fluorescence lifetime is the characteristic time a fluorophore remains in its excited state before returning to the ground state. For a single-exponential decay, the lifetime corresponds to the time at which fluorescence intensity has decreased to approximately 37% of its initial value.
Fluorescence intensity is a relative measurement that can depend on parameters such as fluorophore concentration, excitation power and photostability. Fluorescence lifetime provides complementary information about excited-state dynamics and is less dependent on these intensity-related experimental parameters.
Time-Correlated Single Photon Counting (TCSPC) is a highly sensitive technique for measuring fluorescence lifetimes. It records the arrival times of individual emitted photons relative to repeated excitation pulses and reconstructs the fluorescence decay from the resulting photon-arrival histogram.
A sample is repeatedly excited using a pulsed light source. The arrival time of individual fluorescence photons is measured relative to the excitation pulse. After many excitation cycles, these events are accumulated into a histogram representing the fluorescence decay, which can then be fitted to determine fluorescence lifetimes.
Species with similar or overlapping fluorescence spectra can have different fluorescence lifetimes. Lifetime measurements therefore add a temporal dimension that can help distinguish emitting species that may be difficult to separate using steady-state fluorescence alone.
Time-Resolved Emission Spectroscopy combines spectral and temporal information by monitoring fluorescence emission as a function of both wavelength and time. It can help separate different emitting species and reveal how fluorescence spectra evolve after excitation.
Yes. Fluorescence lifetime analysis can investigate fluorophores and their molecular environments in biological systems. Examples include protein fluorescence, molecular binding, quenching and changes in fluorophore environment.
Long-lived phosphorescence can be investigated using techniques such as Multi-Channel Scaling (MCS), Boxcar detection and Single-Shot Transient Digitization. These approaches extend time-resolved spectroscopy to microsecond, millisecond and even longer emission processes.
